The first is the serial dilution series in which a series of single tubes of test and reference media is each inoculated with standard volumes of diluted inoculum. As will be shown below, this approach lacks precision.

Statistical techniques can be used also to optimise test precision whilst minimising labour and material inputs. The procedures described are predicated on the inoculation of test and reference media using standardised inocula of suitable microorganisms. When assessing the quality of a medium against a previous batch, it is essential to recognise that the quality of a ‘‘reference’’ medium may deteriorate during storage so that gradual loss of quality characteristics may not be recognised. It is therefore essential that laboratories ensure consistency in the preparation and storage of media and inocula and in the evaluation and recording of quality test data.

For smaller rods and cocci, the volume occupied will be smaller, although yeasts and some other microbial forms may be larger (Jarvis, 2008). So it is not unreasonable to assume that the distribution of microbial cells and cell aggregates in a liquid medium is random and therefore conforms to a Poisson distribution. However, cell aggregates (clumps) may become partially or completely disassociated when a suspension of cells is mixed, so affecting the apparent number of cfu. 2 Approximate 95% confidence limits for numbers of colonies assuming agreement with Poisson distribution (Jarvis 2008).
